Analytical Biochemistry

3.  Course Aims & Objectives:

  • To introduce to students the basic principles of techniques used routinely in modern biochemistry and molecular biology.

  • On completion of this course, students should be able to:

    • demonstrate an understanding on the basic principles and current status of analytical biochemistry;
    • critically evaluate recent publications in research and development of analytical biochemistry;
    • apply conventional bioanalytical techniques to chemical and biochemical research when necessary.

    • General principles of biochemical investigation

    In vivo and in vitro studies. Physiological solutions. Cell and tissue culture. Centrifugation.

    • Nucleic acids and proteins

    Structure and functions of nucleic acids. Replication, transcription, and translation. Protein structure. Enzymes.

    • Isotopic and non-isotopic labellings

    Labeling and detection of radioisotopes. Luminescence and chemiluminesence labels. Enzymatic labels.

    • Immunochemical techniques

    General principles. Antibody production. Immunoprecipitation. Immonoassays. ELISA. Immunohistochemistry.

    • Molecular biology techniques

    Synthesis and sequencing of DNA. Enzymatic manipulation of gene. Gene cloning and expression. PCR.

    • Protein and enzyme techniques

    Protein purification. Enzyme kinetics. Enzyme assays. Protein-ligand binding studies. Immobilized enzymes.

    • Chromatographic and electrophoretic techniques

    Adsorption, partition, ion-exchange, size-exclusion, and affinity LC’s. Agarose gel electrophoresis. PAGE. Capillary electrophoresis.

    • Microscopic techniques

    Fluorescence microscope. SEM and TEM. STM and AFM.

    • Spectroscopic and electrochemical techniques

    UV-visible. Spectrofluorimetry. Circular dichroism. MS. NMR. Redox enzymes. Oxygen and pH electrodes. Biosensors.

    Examination duration: 3 H
    Percentage of coursework, examination, etc.:
    Examination 70%
    Course work 30%
    All students must satisfy the following "Minimum Passing Requirement" for BCH courses:
    "A minimum of 30% in coursework as well as in examination, in addition to a minimum of 40% in coursework and examination taken together".
